Research Interests

My research interest use functional genomic approaches to dissect two complex traits of economical and ecological importance in maritime pine (drought tolerance and wood properties).

· 1/ Relevant traits are measured in natural populations (to assess their natural variability) and in experimental designs of the maritime pine breeding program (to determine the extent of genetic control).

· 2/ Genetic maps are constructed, QTLs for these traits are mapped and positional candidate genes (i.e. colocalize with QTLs) identified and compared to the position obtained in a phylogenetically related species : P. taeda.

· 3/ Transcriptomics (EST sequencing, targeted identification of important genes using differential display techniques, expression analysis using high density filters) and proteomics (2D PAGE coupled with protein identification by mass spectrometry) are used in concert to identify expressional candidate genes involved in the studied traits. We have also started experiments to determine the extent to which quantitative phenotypic variance is mirrored by quantitative variation of proteins (PQL mapping).

· 4/ Functional candidate genes are also identified by literature and database search, e.g. homologs of key genes in Arabidopsis are being investigated in pine.

The advent of genomic technologies (high throughput sequencing and genotyping methods) makes it possible to scan the genes for SNP and associate these with traits in natural populations. To efficiently use germplasm diversity for genomics, we first investigated the level, structure and history of diversity using neutral markers (nuclear and chloroplastic microsatellites, AFLP). Second we are conducting experiments to identify the individual nucleotides at candidate genes that are responsible for quantitative traits. Association studies based on linkage disequilibrium should provide high resolution for identifying genes that may conribute to phenotypic variation. We are starting this research from two angles:

· The previous research is suggesting candidate genes to be evaluated with association methods in natural populations.

· We are also starting with interesting sets of genes and exploring their association with candidate traits.

More recently I was also involved in functional genomic projects in oak (bud phenology, drought stress response) and poplar (drought stress response).
